FOXO4-DRI is a first-in-class senolytic peptide designed to selectively eliminate senescent cells, aged cells that have stopped dividing but remain metabolically active, secreting pro-inflammatory factors (the SASP). The peptide works by disrupting the interaction between FOXO4 and p53, two proteins that cooperate to protect senescent cells from apoptosis. By breaking this protective interaction, FOXO4-DRI restores p53's pro-apoptotic function specifically in senescent cells, triggering their selective removal.
Senescent cell accumulation is a hallmark of biological aging, contributing to tissue dysfunction, chronic inflammation, and age-related disease.
